A detailed guide to how Re-Strainer and Pluristrainer works

Filtration of large sample volumes is easy with a Re-Strainer and Pluristrainer. Continue reading to learn more about how both of them work. 

It is essential to correctly prepare the sample in order to achieve a smooth cell flow. The type of cells to be examined, as well as any other potential impediments to the flow, must be considered.

Cell strainers are sterile, user-friendly devices used to obtain uniform cell suspension from tissues. It is used as a more convenient and faster alternative to gauze filtration. They are ideal for preparing flow cytometry samples and filtering agglutinative proteins generated during serum inactivation.

Our best Lab Cell Strainers are made of a strong nylon mesh with evenly spaced mesh pores. They are sterile, simple devices for quickly isolating primary cells and obtaining a uniform single-cell suspension from tissues. Designed to protect expensive flow cytometry and cell sorting equipment by removing clumps and debris from cell suspensions and clinical samples prior to analysis.

How Do The Cell Strainers Work?

Cell strainers separate debris or clumps from samples, allowing cells to be isolated for further examination. To create a tight fit, the cell strainers fit on the tops of various size sample tubes, so the material must pass through the strainer.

Only certain materials are permitted to pass through the mesh, which is made up of varying sizes. It enables material separation based on size, allowing for the isolation of specific material from the rest of the sample.

What is a Re-Strainer?

The Re-Strainer is a high-volume liquid filtration device that can exclude or concentrate particles from liquids while also allowing for particle recovery. Unscrew the cap to gain access to the 6 ml recovery reservoir. There are two female Luer-lock ports on the Re-Strainer.

These ports can be used to connect multiple Re-Strainers to a filter cascade (male to male Luer-lock connector) or to incorporate the Re-Strainer as an in-line-strainer using a Luer-lock tube adaptor.

The mesh is an integral part of the housing, providing maximum mechanical stability to the device.

What is a PluriStrainer?

The pluriStrainer is a sieving device (or cell strainer) used in a variety of laboratory applications that require liquid filtration and purification. Its one-of-a-kind design includes improved ventilation to prevent clogging for smart sample preparation. PluriStrainer can be stacked to allow direct filtration with various mesh sizes and can also be inverted to recover the sieved material. 

It can be combined with a funnel for larger sample volumes. It is compatible with all major brand 50 mL centrifuge (conical) tubes. The combination of the pluriStrainer and the Connector Ring enables the use of low pressure, which aids in filtration. 

Furthermore, the liquid on top of the mesh can be held back by blocking the flow through the sieve. The liquid on top of the mesh can be used for sample preparation and incubation, such as tissue dissociation or cell lysis (e.g., Buffer RLT, TRIzol).

When should you use Pluristrainer?
  • The actual single-cell suspension is produced following the enzymatic digestion of mammary tissue and organoids.
  • Physical dissociation of the primary tissue
  • Single-cell suspensions of blood cells from the thymus, pancreas, lymph nodes, bone marrow, and other sources are prepared.
  • Preparation of a real single-cell suspension for flow cytometry 
  • Using gauze for filtering is a quick and simple solution.
  • Incubation of cells/cytokines for a short period of time (in combination with pluriBead and Connector Ring)
  • Complex and viscous liquids, such as fat tissue, are sieved (in combination with a low-pressure syringe and a Connector Ring).
What are the applications of PluriStrainer?
PluriStrainer

Attach the 50 mL tube to the pluriStrainer.

On top of that, place your sample material. Your sample material will only be processed in part. By inserting a syringe into the Connector Ring, you can force low pressure to support the straining of rough sample material while pulling the piston.

Stacked pluriStrainer

PluriStrainers are available in a variety of mesh sizes that can be stacked for cascade straining of multiple cell sizes at the same time. 

Reversed pluriStrainer

To obtain the larger fraction, remove the pluriStrainer, turn it upside down on another 50 ml tube, and flush the sample back into the pluriStrainer. 

PluriStrainer with Connector Ring 

It lets you control the flow rate by opening and closing the Luer-Lock, which is useful for physical tissue dissociation (brain, spleen, etc.). 

The sample material will flow through when you open the Luer-Lock. 

PluriStrainer with Funnel

Insert a funnel into the pluriStrainer (42-50000-03). 

You can also add up to 24 mL of sample material.

Connector Ring, PluriStrainer, and Syringe

By attaching a syringe to the Connector Ring, you can apply low pressure to aid in the straining of rough sample material while pulling the piston.
