TwinSpin centrifugation tubes, pre-filled with Density Gradient Medium (DGM), are utilized for optimal separation of peripheral blood mononuclear cells from whole blood and bone marrow. Consisting of a standard 15 ml tube and an inner tube, the inner tube has an open bottom submerged in the DGM. Diluted anti-coagulated blood or bone marrow can be easily pipetted into the TwinSpin, with the sample resting on top of the DGM inside the inner tube. During centrifugation, leukocytes, lymphocytes, and PBMCs separate from unwanted erythrocytes and granulocytes based on the DGM. This results in an enriched interphase containing target cells above the DGM, while erythrocytes sediment through the density gradient media out of the inner tube at the bottom of the outer tube. After separation, the inner tube can be removed, and the elastic cap functions as a valve. By pushing the cap down, the collection tube converts into a pipette, allowing the contents to be collected drop by drop.
