TwinSpin centrifugation tubes, filled with Density Gradient Medium (DGM), offer an effective method for separating peripheral blood mononuclear cells from whole blood and bone marrow. The TwinSpin consists of a standard 15 ml tube with an inner tube. The inner tube has an open bottom and is submerged in the DGM. To use the TwinSpin, diluted anticoagulated blood or bone marrow can be easily pipetted into it, allowing the sample to rest on top of the DGM inside the inner tube.

During centrifugation, leukocytes, lymphocytes, and PBMCs separate from unwanted erythrocytes and granulocytes, based on the DGM. This results in an enriched interphase of target cells above the DGM, while erythrocytes sediment through the density gradient media out of the inner tube at the bottom of the outer tube. Once separation is complete, the inner tube can be removed. The elastic cap serves as a valve, allowing the collection tube to function as a pipette. The contents can then be collected drop by drop.
